The most direct route is a drive through Mombasa Island along the Likoni Ferry and south down the shoreline through Likoni, Tiwi and finally to Diani. In a few months, a new route to South Coast through Dongo Kundu road will open up and create an alternative way to get to Diani Beach.
Getting to Diani Beach without a car is totally doable! There are shared buses that will take you from Airport to Mombas town for cheap. The routes might be kind of confusing though, as it is not a direct route. From Mombasa, take another shared van (matatu) to Likoni and cross the ferry on foot.
Once across the Likoni side, find another Matatu to Ukunda – the township next to Diani. Alight there and flag down a tuk-tuk! It’s a 3 wheeled vehicle that fits about 3 people and are a fun way to get around locally.
